Janus kinase 1 (JAK1) is an intracellular enzyme that partners with other Janus kinases at the inner surface of cell-membrane receptors for many cytokines—signaling proteins such as interleukin-6, interleukin-4, interferons, and others involved in immune activation. When a cytokine binds its receptor, JAK1 becomes active and phosphorylates STAT proteins, which then travel to the nucleus and switch on genes that drive inflammation and immune cell proliferation. This cascade is called JAK-STAT signaling. Because JAK1 sits downstream of so many inflammatory cytokine receptors, blocking it with a small-molecule inhibitor dampens multiple inflammatory signals at once rather than neutralizing a single cytokine. Selectivity for JAK1 over the related kinases JAK2, JAK3, and TYK2 is a deliberate design goal: it aims to preserve efficacy against inflammatory cytokines while reducing interference with JAK2-dependent blood cell production and JAK3-dependent lymphocyte development, which are associated with anemia, low blood counts, and broader immune suppression when disrupted. This mechanism is relevant across autoimmune and inflammatory diseases—conditions affecting the joints, skin, gut, and other tissues—where excessive cytokine signaling through the JAK-STAT pathway sustains chronic inflammation. Oral JAK1 inhibitors offer a convenient alternative to injectable biologic therapies that target single cytokines.
