JAK1 and JAK3 are members of the Janus kinase family, enzymes that sit inside the cell just beneath many cytokine receptors on the cell surface. When a cytokine binds its receptor, JAK enzymes become active and add phosphate groups to STAT proteins, which then travel to the nucleus and switch on genes involved in immune cell activation and proliferation. JAK1 and JAK3 partner together specifically downstream of receptors that use a shared subunit called the common gamma chain, through which cytokines such as interleukin-2, -4, -7, -9, -15, and -21 drive T-cell, natural killer-cell, and other lymphocyte responses. These pathways are central to many inflammatory and autoimmune skin conditions, where excessive cytokine signaling sustains itching, redness, and tissue damage. Inhibiting JAK1/JAK3 dampens this signaling cascade at its source rather than blocking a single cytokine, giving broad anti-inflammatory activity. Delivering such an inhibitor as a topical, 'soft' formulation means the drug is designed to act mainly within the skin, with limited entry into the bloodstream, aiming to preserve local efficacy while reducing the systemic exposure associated with oral JAK inhibitors. This approach is broadly relevant to chronic inflammatory dermatologic diseases where localized cytokine-driven inflammation predominates.
