This mechanism targets two molecules involved in type 2 inflammation, a pattern of immune activity implicated in asthma, atopic dermatitis, and related allergic conditions. Thymic stromal lymphopoietin (TSLP) is an alarmin, a signaling protein released by epithelial cells lining the airways or skin when they are irritated by allergens, pollutants, or infection. TSLP acts high in the inflammatory cascade, activating dendritic cells and innate lymphoid cells that in turn drive production of downstream effector cytokines, one of which is interleukin-13 (IL-13). IL-13 acts closer to the tissue, promoting mucus overproduction, airway narrowing, and structural remodeling. A bispecific antibody that neutralizes both molecules simultaneously aims to interrupt the pathway at two points: near its origin (TSLP) and at its point of tissue damage (IL-13). This dual approach is intended to produce more complete suppression of type 2 inflammation than blocking either molecule alone, since IL-13 production can be sustained through routes that do not strictly depend on TSLP. The broad relevance of this mechanism lies in diseases where epithelial-driven inflammation and its downstream effector signaling both contribute meaningfully to symptoms and tissue damage.
