TSLP (thymic stromal lymphopoietin) is a cytokine released by the epithelial linings of the skin, airway, and gut when they sense allergens, irritants, or pathogens. It acts near the top of the type 2 immune cascade, activating dendritic cells, innate lymphoid cells, and T helper 2 cells that in turn produce interleukins driving allergic inflammation. Separately, stem cell factor (SCF) signaling through its receptor KIT is required for the development, survival, and activation of mast cells, the cells that release histamine and other mediators responsible for itching, swelling, and airway narrowing. A bispecific antibody that neutralizes TSLP while also blocking SCF-KIT signaling combines two complementary actions: quieting the epithelial alarm signal that initiates broad type 2 inflammation, and directly reducing the number and reactivity of mast cells, an effector population that TSLP blockade alone does not fully control. This dual mechanism is designed for conditions where both upstream inflammatory signaling and mast-cell-driven symptoms contribute, such as chronic urticaria, severe asthma, atopic dermatitis, and mast cell disorders. By addressing both the initiating and effector arms of the response, the approach aims for more complete disease control than targeting either pathway alone.
