Stem cell factor (SCF), also known as KIT ligand, is a signaling protein that binds to the receptor KIT on the surface of mast cells, a type of immune cell involved in allergic and inflammatory responses. Engagement of KIT by SCF promotes mast cell survival, proliferation, and activation, leading these cells to release histamine and other mediators that drive itching, redness, and inflammation. In chronic inflammatory skin conditions such as atopic dermatitis, mast cells accumulate and become persistently activated, contributing to the itch-scratch cycle and skin barrier disruption that characterize the disease. Monoclonal antibodies designed to neutralize circulating SCF act by intercepting the ligand before it can engage KIT, thereby reducing the pool of activated mast cells and dampening downstream inflammatory signaling. This approach targets a root driver of mast cell biology rather than blocking a single downstream mediator, offering a rationale for addressing itch and inflammation across mast-cell-driven skin diseases. Because SCF-KIT signaling also supports other cell populations, including hematopoietic stem cells, melanocytes, and germ cells, therapeutic modulation of this axis requires careful consideration of effects beyond the skin.
