Evidence outlook · lead readout
NCT05424276 · Parkinson Disease
The completed lead evidence is positive, with design limits that temper confidence — see the rationale below for exactly which.
NCT05424276: The trial was well designed - randomized, blinded, and placebo-controlled with roughly 33-35 patients per arm - but its designated primary endpoint was adverse-event incidence rather than a disease-related clinical outcome, and no statistical comparison across arms was reported for that primary endpoint (event rates were similar across all four groups: 54-64%). The efficacy-related measures (MDS-UPDRS, PDQ-39, etc.) were only secondary and the study was explicitly stated as not powered to detect efficacy differences, with results showing no consistent separation from placebo.
