AMBITION (NCT01178073): This large, randomized, quadruple-blind trial compared first-line combination ambrisentan plus tadalafil against monotherapy with either drug in patients newly diagnosed with pulmonary arterial hypertension, using a hard clinical composite endpoint (time to a first clinical failure event, including death, hospitalization, and disease progression). Combination therapy significantly cut the risk of clinical failure compared with monotherapy (hazard ratio 0.502, p=0.0002), supporting ambrisentan's use as part of first-line combination treatment.
