A randomized, quadruple-blind, placebo-controlled phase 3 trial enrolling 387 participants with moderate-to-severe atopic dermatitis. The co-primary endpoints (IGA response and EASI-75 at week 12) were both statistically significant for the 100 mg and 200 mg doses versus placebo, with large absolute differences (e.g. 43.8% vs 7.9% for IGA). The adequate sample size, blinded design, and positive hard clinical endpoints justify the strong evidence grade.

Participants were randomized and the trial was quadruple-blinded and placebo-controlled in 912 adolescents and adults. Both co-primary endpoints -- a 75% reduction in the validated Eczema Area and Severity Index (EASI75) and clearance or near-clearance on the validated Investigator Global Assessment -- were clearly met for both upadacitinib doses versus placebo at week 16 (p<0.001 for all comparisons).
