Janus kinases (JAK1, JAK2, and JAK3) are enzymes that sit just inside the cell membrane, attached to receptors for cytokines—small signaling proteins that regulate immune and blood cell activity. When a cytokine binds its receptor, associated JAK enzymes activate and phosphorylate STAT proteins, which then travel to the nucleus and switch on genes controlling cell growth, inflammation, and blood cell production. Different cytokines rely on different combinations of the three JAK enzymes, so a molecule that inhibits all three, rather than a single subtype, can dampen signaling from a broad range of cytokines at once. This breadth is useful in diseases driven by multiple overlapping inflammatory or proliferative signals, such as autoimmune and inflammatory conditions, and certain blood disorders where abnormal JAK signaling drives excessive cell proliferation. By blocking the kinase activity of JAK1/2/3, an oral small molecule inhibitor reduces downstream STAT activation and the resulting gene expression programs, lowering inflammatory output or curbing abnormal cell growth. Because JAK enzymes are used broadly throughout the immune and hematologic systems, their inhibition is a widely applicable but non-specific strategy, with therapeutic relevance spanning several distinct disease categories rather than a single narrow indication.
