This combination pairs two nuclear hormone receptors that govern skin cell behavior through different but complementary mechanisms. The glucocorticoid receptor, once activated by a corticosteroid, moves into the cell nucleus and alters gene expression to suppress inflammatory signaling, dampening the immune activity that drives redness, swelling, and itching in inflammatory skin diseases. The retinoic acid receptor, activated by a vitamin A-derived retinoid, regulates genes controlling how skin cells mature and multiply, correcting the abnormal, excessive proliferation and faulty differentiation of keratinocytes (the main cells of the epidermis) seen in conditions such as psoriasis. Combining a corticosteroid with a retinoid in one formulation addresses both problems simultaneously: the corticosteroid controls inflammation, while the retinoid normalizes the thickened, scaling skin architecture that inflammation alone does not fully resolve. This dual approach can also allow lower doses of each component than would be needed if either were used alone, which is relevant because both drug classes carry dose-related skin tolerability concerns. This receptor pairing is broadly relevant to chronic inflammatory and hyperproliferative skin disorders where both immune dysregulation and abnormal epidermal turnover coexist.
