Dopamine D2 and D3 receptors, along with the serotonin 5-HT7 receptor, are G protein-coupled receptors relevant to psychiatric disease, particularly schizophrenia and mood disorders. D2 and D3 receptors normally dampen neuronal signaling when activated by dopamine; blocking them reduces the excess dopaminergic transmission in certain brain circuits that is thought to underlie hallucinations and delusions. D3 receptors are concentrated in limbic brain regions involved in emotion and reward, so targeting D3 alongside D2 may address symptoms tied to motivation and mood with a different side-effect profile than D2 blockade alone. The 5-HT7 receptor operates through a distinct signaling route and has been linked to circadian rhythm regulation, mood, and cognition. Adding 5-HT7 antagonism to D2/D3 blockade is intended to temper some of the drawbacks of dopamine blockade while contributing independent benefits for mood and cognitive symptoms, which are often poorly addressed by dopamine-focused treatment alone. This combined receptor profile reflects a broader trend in psychiatric drug design: pairing dopamine antagonism, which addresses core psychotic symptoms, with serotonergic modulation aimed at negative symptoms, cognitive deficits, or comorbid depression, conditions that frequently accompany psychotic disorders and are not well served by older, purely dopamine-blocking agents.
