Dopamine D2 and D3 receptors, along with the serotonin 5-HT1A receptor, belong to families of G-protein-coupled receptors that regulate mood, motivation, cognition, and movement across several brain circuits. In psychotic and mood disorders, dopamine signaling through D2/D3 receptors is thought to be excessive in some brain regions and insufficient in others, while serotonergic tone through 5-HT1A contributes to anxiety and mood regulation. Rather than simply blocking dopamine receptors, as older antipsychotics do, this mechanism uses partial agonism: the compound occupies D2/D3 receptors and produces a fraction of dopamine's own effect, effectively dampening signaling where dopamine is overactive while providing a baseline of stimulation where it is underactive. Combined partial agonism at 5-HT1A adds an anxiolytic and mood-stabilizing influence and can further modulate dopamine release in cortical regions. This dual receptor profile is used across schizophrenia, bipolar disorder, and as an adjunct in major depressive disorder, where the goal is to normalize dysregulated dopamine and serotonin activity rather than uniformly suppress it. The approach is intended to treat core symptoms while reducing the risk of certain movement-related and hormonal side effects associated with full dopamine receptor blockade.
