This mechanism targets dopamine D2 and D3 receptors, with a preference for the D3 subtype, using a partial agonist rather than a full blocker or activator. A partial agonist binds the receptor but produces a submaximal response, so it can either dampen excessive dopamine signaling or supply modest stimulation where dopamine tone is low, functioning as a stabilizer of dopaminergic activity rather than a simple on/off switch. This profile is combined with partial agonism at serotonin 5-HT1A receptors and antagonism at 5-HT2A (and related 5-HT2B) receptors, an activity pattern shared by several atypical antipsychotics that is thought to soften motor side effects and provide benefit for mood and negative symptoms alongside psychosis control. The rationale for emphasizing D3 selectivity is that this receptor subtype is concentrated in limbic brain regions linked to motivation, reward, and cognition, so preferential engagement there may address symptoms poorly served by older, D2-dominant antipsychotics. Such agents are studied broadly in schizophrenia and related psychotic and mood disorders, where restoring balanced dopaminergic and serotonergic signaling, rather than uniformly suppressing it, is the guiding therapeutic principle.
