Participants were randomized to donanemab or active-control aducanumab, but the trial was open-label rather than blinded. Both prespecified primary amyloid-clearance comparisons favored donanemab with statistically significant results; the biomarker-based primary endpoint and lack of blinding support a moderate evidence grade rather than a strong one.

Randomized, triple-masked phase 2b in early Alzheimer's disease (856 participants). The Bayesian primary analysis of the ADCOMS composite at 12 months estimated a 63.8% posterior probability of meaningful benefit for the highest dose, short of the study's prespecified success criterion, so the primary endpoint was not met.
