TRPV1 (transient receptor potential vanilloid 1) is a nonselective cation channel found mainly on peripheral sensory nerve fibers, particularly the small unmyelinated C-fibers and thinly myelinated A-delta fibers that carry pain and temperature signals from the skin. It is the molecular receptor for capsaicin, the pungent compound in chili peppers, and it is also opened by noxious heat and by the acidic conditions that accompany tissue injury, so it functions as a converging sensor for chemical, thermal, and inflammatory pain signals.

Pharmacologically, TRPV1 agonists exploit a two-phase response: an initial burst of channel opening produces a transient sensation of burning or warmth, but repeated or sustained exposure drives the nerve fibers into a desensitized state, sometimes called defunctionalization, in which their capacity to transmit pain signals is markedly reduced for a period of time. This receptor-driven stimulate-then-quiet effect is the pharmacological basis for using topical vanilloid compounds as external analgesics.

This mechanism is broadly relevant to conditions dominated by localized musculoskeletal or neuropathic pain, where reducing the excitability of superficial sensory nerve endings can ease chronic discomfort without acting on central nervous system pathways such as opioid or GABA receptors, making it a useful complement to systemic pain medications.
