TRPV1 (transient receptor potential vanilloid 1) is an ion channel found on sensory nerve endings throughout the skin, airways, bladder, and gut. It normally opens in response to heat, acidic environments, and irritant chemicals such as capsaicin, allowing calcium and sodium ions to flow into the nerve cell and trigger a signal that the brain interprets as pain, burning, or itch. In many chronic conditions, this channel becomes persistently overactive, contributing to ongoing pain, neurogenic inflammation, and itch even without an obvious external trigger. A TRPV1 antagonist is a molecule that binds the channel and prevents it from opening, dampening this abnormal sensory signaling at its source rather than blocking pain perception centrally. This mechanism has been explored broadly across chronic pain syndromes, neuropathic pain, certain inflammatory skin conditions, and other disorders where excessive activation of sensory nerves drives symptoms. Because TRPV1 is expressed on peripheral nerve terminals, antagonists can act locally or systemically depending on formulation and distribution, offering a pathway-specific alternative to broader analgesics such as opioids. The channel's central role in temperature sensing also means its modulation intersects with body temperature regulation, which is a defining biological feature of this target class.
