Toll-like receptor 7 (TLR7) is a pattern-recognition receptor found mainly inside endosomes of immune cells such as plasmacytoid dendritic cells, monocytes, and B cells. Its natural role is to detect single-stranded RNA characteristic of viruses, triggering an innate antiviral response. TLR7 agonists are synthetic small molecules designed to bind this receptor and switch on the same signaling cascade in the absence of actual infection, prompting cells to release type I interferons and other cytokines that recruit and activate broader immune defenses, including natural killer cells and cytotoxic T cells. The biological rationale for pharmacologically engaging TLR7 is to harness this innate-immune amplification for conditions where the body's own antiviral or antitumor surveillance is insufficient, including chronic viral infections and certain cancers, where enhanced interferon signaling and immune cell activation may help control or clear disease. Because TLR7 sits upstream of a network of adaptive immune responses, agonism is generally considered a strategy to prime or reawaken immune engagement rather than an agent with disease-specific action; it works by broadly stimulating immune vigilance and depends on which target-directed responses are able to unfold downstream.
