TLR7 and TLR8 are toll-like receptors, pattern-recognition proteins that reside in the endosomal membranes of immune cells such as dendritic cells, monocytes, and B cells. Their natural role is to detect single-stranded RNA, a molecular signature common to many viruses, and to trigger the innate immune system's alarm response. An agonist is a molecule that activates a receptor in place of, or alongside, its natural trigger. A TLR7/8 agonist mimics viral RNA closely enough to switch on this same alarm pathway deliberately, prompting immune cells to produce type I interferons and pro-inflammatory cytokines and to mature into more effective antigen-presenting cells. This makes TLR7/8 agonism relevant wherever a stronger, better-directed immune response is therapeutically desirable: as an adjuvant that sharpens the response to a vaccine antigen, as a topical treatment for virally driven or superficial skin lesions, and, increasingly, as a way to convert tumors that evade immune detection into ones that provoke an active anti-tumor response, often by injecting the agonist directly into a lesion to limit systemic exposure. The central biological rationale is the same across these settings: harnessing an ancient antiviral sensing pathway to recruit and instruct the immune system where it is otherwise insufficiently engaged.
