The kappa opioid receptor (KOR) is a cell-surface receptor belonging to the G protein–coupled receptor family, normally activated by its endogenous ligand dynorphin. Unlike the mu opioid receptor, which mediates pain relief and euphoria, KOR activation tends to produce dysphoria (a state of unease or low mood) and blunts reward signaling in brain circuits that use the neurotransmitter dopamine. This makes the dynorphin–KOR system a key biological brake on mood and motivation, particularly during stress. Blocking KOR with an antagonist removes this brake, and is therefore explored as a strategy to relieve anhedonia (loss of interest or pleasure) and low mood seen in depressive disorders, and to reduce stress-triggered urges to use addictive substances. Because KOR antagonism acts on a modulatory pathway rather than replacing a neurotransmitter directly, its effects are expected to be more subtle and better tolerated than broader dopamine-stimulating approaches. This mechanism is being studied primarily in psychiatric contexts such as major depressive disorder, particularly for symptoms of anhedonia, and in substance use disorders, where dynorphin-KOR signaling is implicated in stress-induced relapse.
