CD30 ligand (CD30L, also called CD153) is a member of the tumor necrosis factor (TNF) superfamily of proteins, expressed on the surface of activated T cells, B cells, and various myeloid cells. It binds its receptor, CD30, delivering a costimulatory signal — a secondary cue, beyond antigen recognition, that intensifies and shapes an ongoing immune response. This CD30-CD30L axis is best known for driving proliferation and cytokine production in T-cell subsets associated with type-2 and type-17 inflammatory responses, and its receptor is also a marker on malignant cells in certain lymphomas. Modulating this pathway is of interest because excessive or inappropriate CD30-CD30L engagement is implicated in chronic inflammatory and autoimmune skin and tissue conditions, where dampening costimulation could reduce disease activity without broadly suppressing the entire immune system. An antibody directed against the ligand itself, rather than the receptor, works by preventing CD30L from engaging CD30-bearing cells in the first place, blocking the signal at its source. This approach is distinct from strategies that use the receptor as a docking point to deliver cytotoxic payloads to malignant cells; here the goal is to quiet signaling, not to kill cells.
