CD30 is a cell-surface receptor belonging to the tumor necrosis factor receptor superfamily. Under normal conditions, it is expressed at low levels, mainly on activated lymphocytes, but certain lymphomas display it abundantly on the malignant cell surface, most notably Hodgkin lymphoma and some T-cell lymphomas. This restricted, high-density expression pattern makes CD30 an attractive target for an antibody-drug conjugate (ADC), a therapeutic modality combining a targeting antibody with a cytotoxic chemical payload joined by a chemical linker. The antibody component recognizes and binds CD30 on the tumor cell surface, after which the complex is internalized. Inside the cell, the linker is cleaved, releasing the payload, typically a potent microtubule-disrupting agent, directly within the malignant cell. This localized delivery allows use of a cytotoxic potency far exceeding what could be tolerated as a conventional systemic chemotherapy, while sparing many normal tissues that lack the target. The broad rationale for this mechanism is to concentrate cytotoxic activity in CD30-expressing malignant cells across lymphoma subtypes that depend on this marker, improving the therapeutic margin relative to non-targeted cytotoxic chemotherapy.
