Bitter taste receptors, known as TAS2Rs, are a family of G-protein-coupled receptors originally characterized on the tongue, where they detect bitter-tasting compounds as a protective mechanism against ingesting toxins. The same receptor family is also expressed on enteroendocrine cells lining the gut, distant from any conscious sense of taste. Activating these gut-based receptors triggers intracellular signaling that leads to release of hormones such as cholecystokinin, glucagon-like peptide-1, and peptide YY, which are involved in regulating appetite, gastric emptying, and glucose handling. Because these hormones are also targets of established metabolic therapies, agonism of gut bitter taste receptors is of interest as a way to stimulate the body's own hormone release using an orally administered small molecule, rather than delivering the hormone directly. This mechanism is broadly relevant to conditions involving appetite dysregulation, obesity, and impaired glucose control, where boosting endogenous gut-hormone signaling could complement or substitute for other metabolic interventions. The approach is distinguished by acting locally within the gastrointestinal tract rather than systemically, which may shape both its therapeutic profile and its side-effect considerations.
