Amylin receptor agonist

The amylin receptor is a G protein-coupled receptor formed by the calcitonin receptor combined with an accessory protein (a receptor activity-modifying protein, or RAMP). It is the target of amylin, a hormone co-released with insulin from pancreatic beta cells after meals. Amylin acts primarily in the brainstem, particularly the area postrema, a region that senses circulating signals to regulate appetite and gastric function. Activating this receptor slows gastric emptying, promotes a sense of fullness, and dampens glucagon secretion, complementing insulin's role in glucose control. Because amylin signaling is deficient or diminished in diabetes and because appetite suppression through this pathway is distinct from, but complementary to, incretin hormone pathways such as GLP-1, amylin receptor agonism has become relevant to both metabolic disease and weight management. Many agents in development are amylin analogs or engineered peptides designed to resist degradation and extend duration of action, sometimes combined with GLP-1 receptor agonists to achieve additive effects on food intake and body weight through parallel but distinct neural and gastrointestinal mechanisms. This mechanism broadly matters in obesity, type 2 diabetes, and conditions where appetite regulation and glycemic control intersect.
