Dendritic cells are antigen-presenting cells that orchestrate adaptive immune responses by capturing molecular fragments, called antigens, and displaying them on their surface via major histocompatibility complex (MHC) molecules, thereby activating T lymphocytes capable of recognizing and destroying cells bearing those antigens. In cancer immunotherapy, dendritic cell vaccines aim to prime a patient's immune system against antigens characteristic of malignant cells, with the goal of generating durable, tumor-specific immunity. This approach uses an allogeneic, meaning donor-derived rather than patient-derived, dendritic cell line instead of dendritic cells grown individually from each patient's own blood. The cell line naturally expresses a defined set of leukemia-associated antigens, so a single standardized product can be manufactured at scale and given across patients whose leukemic cells share these markers, rather than requiring bespoke preparation for every individual. The underlying pathway is anti-tumor immune activation through antigen presentation: vaccine cells display leukemia antigens on MHC molecules, are engaged by the patient's immune system, and stimulate expansion of antigen-specific T cells able to target residual leukemic cells. This mechanism is broadly relevant to hematologic malignancies, particularly myeloid leukemias, where deepening and maintaining remission after initial treatment, and controlling minimal residual disease through immune surveillance, remain central clinical goals.
