ALK7 (activin receptor-like kinase 7, also called ACVR1C) is a type I serine/threonine kinase receptor belonging to the transforming growth factor-beta (TGF-β) superfamily. It is expressed prominently in adipose tissue and pancreatic beta cells, where it responds to ligands such as activin B and GDF3. Engagement of ALK7 activates intracellular SMAD2/3 signaling, which in adipocytes tends to restrain lipid breakdown (lipolysis) and influence how fat cells store energy, and in beta cells affects insulin secretion. Because of this role, ALK7 has been studied as a target for metabolic disease: reducing its signaling in fat tissue is associated, in preclinical models, with increased fat mobilization and improved insulin sensitivity. Silencing ALK7 expression with small interfering RNA (siRNA) represents one strategy to achieve sustained reduction of receptor levels rather than transiently blocking receptor activity with a small molecule or antibody. This mechanism is broadly relevant to obesity, insulin resistance, and related metabolic disorders, where altering energy storage and glucose-handling pathways is a therapeutic goal.
