5T4, also called trophoblast glycoprotein (TPBG), is a cell-surface protein that is abundant during early embryonic development but largely absent from normal adult tissues, while being re-expressed at high levels on many solid tumors. This pattern of restricted, tumor-associated expression makes it an attractive address for targeted therapy. The mechanism described here uses an antibody directed against 5T4 not to block a signaling function directly, but as a delivery vehicle: the antibody binds 5T4 on the tumor cell surface, the complex is drawn inside the cell (internalized), and a potent cytotoxic payload—monomethyl auristatin E (MMAE), a microtubule-disrupting agent—is released within the cell to kill it. This antibody-drug conjugate (ADC) strategy aims to concentrate a chemotherapy-like agent selectively in tumor cells while sparing normal tissue that lacks the target, in principle widening the therapeutic window compared with conventional systemic chemotherapy. Because 5T4 is expressed across a range of epithelial cancer types, this approach is relevant broadly in oncology wherever tumors upregulate this oncofetal antigen. The rationale rests on combining tumor-selective binding with a payload potent enough to kill cells even at the relatively low intracellular concentrations achievable through receptor-mediated uptake.
