Nivolumab vs Sintilimab

Same mechanism, same race: design-graded trial evidence for both assets, with the grading rationale shown. Grades describe the quality of the evidence, never the merits of either security.

Nivolumab

Marketed by Bristol-Myers Squibb

Approved in Non-Small Cell Lung Cancermonoclonal antibody
Partial signal

NCT01673867 · Phase 3 · completed

This was a large randomized trial comparing nivolumab with active treatment, but it was open-label rather than double-blinded. The prespecified primary endpoint, overall survival, favored nivolumab and was statistically significant; however, the lack of blinding limits the strength of the evidence.

Sintilimab

Developed by Innovent Biologics and partners

Phase 3 in Non-Small Cell Lung Cancermonoclonal antibody
Partial signal

NCT03607539 · Phase 3 · completed

This trial was randomized, quadruple-blinded, and placebo-controlled with a reasonably large sample (397 patients) and used a hard clinical primary endpoint (progression-free survival), which is a strong design overall. However, the results record reports only median PFS values and confidence intervals for each arm (8.9 vs 5.0 months) without a p-value or hazard ratio, so whether the primary comparison reached statistical significance cannot be confirmed from the data provided.
