The study randomized 346 participants to double-blind 56 mg esketamine, 84 mg esketamine, or matching placebo, each with an oral antidepressant, and used the clinical MADRS depression score as its primary endpoint. The 56 mg dose significantly outperformed placebo, while the 84 mg dose did not, so the primary results were mixed despite the strong trial design.

Zelquistinel

The study randomized 226 participants overall to several zelquistinel dose groups or placebo and masked both participants and investigators, but it stated that no power calculation was performed and each comparison was relatively small. The primary MADRS comparisons were clinical measures but were missed, with all reported p-values above 0.05, so the findings provide limited evidence of benefit.
