This was a randomized, triple-blind (participant, care provider, investigator), double-dummy, active-controlled phase 3 trial in 724 treatment-experienced HIV patients, comparing elvitegravir with raltegravir, both given with an optimized background regimen. The primary endpoint, confirmed HIV-1 RNA <50 copies/mL at week 48, was met as a non-inferiority result: 59.0% with elvitegravir vs 57.8% with raltegravir, with the 95% confidence interval for the difference (-6.0 to 8.2) falling entirely within the prespecified 10% margin. The trial was adequately powered (planned 700 participants, 85% power for non-inferiority).

The study was randomized and double-blind with matching placebo, but it was very small, with 6, 6, and 7 participants receiving VH4524184 doses and only 3 receiving placebo. Its primary endpoint was a surrogate measure of plasma HIV-1 RNA change, and although the reported mean changes favored the medium and high doses, statistical significance was not reported.
