The study randomized 146 participants overall, including 130 in the main Part B comparison, to double-blind CLE-100 or placebo alongside standard antidepressant treatment. Its clinical MADRS primary endpoint was missed: the improvement with CLE-100 was not significantly different from placebo (p=0.46), so the trial provides well-controlled but inconclusive evidence.

Participants were randomized and blinded to MIJ821, placebo, or ketamine, but the 70-person study was small and spread across six treatment groups. The clinical primary endpoint was met at 24 hours for both pooled MIJ821 dose groups versus placebo, with statistically significant MADRS score differences, but the limited sample size supports moderate rather than strong evidence.
