Aflibercept · Eylea
Marketed by Regeneron Pharmaceuticals and partners
NCT00509795 · Phase 3 · completed
The trial randomly assigned 1,217 participants and used quadruple blinding with ranibizumab as an active comparator. Its hard clinical primary endpoint—maintaining vision at week 52—met the prespecified non-inferiority criterion for all three aflibercept regimens, supporting a strong evidence grade.

Pegaptanib (Macugen)
Marketed by Pfizer
NCT01100307 · Phase 3 · completed
This was a randomized, double-masked, sham-controlled phase 3 trial with 243 participants and a hard clinical visual-acuity endpoint. The primary endpoint was met: 25 of 123 participants receiving pegaptanib versus 6 of 120 receiving sham achieved at least a 10-letter improvement at week 24 (p=0.0003).
