Type I interferon receptor subunit 1 (IFNAR1) is one half of a two-part receptor complex that sits on the surface of most cells and detects type I interferons, a family of signaling proteins that includes interferon-alpha and interferon-beta. These interferons are produced during viral infection and also, inappropriately, in certain autoimmune conditions, where sustained interferon activity contributes to tissue inflammation and damage. When interferon binds the IFNAR1/IFNAR2 complex, it triggers an intracellular signaling cascade that switches on hundreds of genes involved in antiviral defense and immune activation. In diseases such as systemic lupus erythematosus, many patients show a persistently elevated interferon signature, and dampening this signaling has been pursued as a therapeutic strategy. Monoclonal antibodies directed against IFNAR1 work by binding this receptor subunit and preventing the multiple type I interferon ligands from engaging it, thereby blunting downstream signaling broadly rather than neutralizing individual interferon molecules one at a time. This approach is relevant across autoimmune and inflammatory conditions where chronic type I interferon activity is thought to drive pathology, while also carrying an inherent trade-off, since the same pathway is central to normal antiviral immunity.
