TNFR2 (tumor necrosis factor receptor 2) is a cell-surface receptor for the cytokine TNF-alpha, expressed mainly on immune cells such as regulatory and effector T cells, natural killer (NK) cells, and endothelium. Unlike its sibling receptor TNFR1, TNFR2 lacks an intracellular death domain and instead signals through adaptor proteins (TRAFs) that activate survival and proliferation pathways, including NF-kB. Because TNFR2 engagement can costimulate CD4 and CD8 T cells and NK cells, agonist antibodies directed at this receptor are being explored as a way to amplify antitumor immune responses, complementing checkpoint blockade or other immunotherapies. The biological rationale is that many tumors evade immune attack partly through insufficient activation of effector lymphocytes; providing an additional costimulatory signal through TNFR2 may increase their expansion, cytotoxic function, and persistence. This mechanism is being explored broadly in oncology, particularly in settings where the tumor microenvironment is immunologically "cold" and effector cell activity is limited. Because TNFR2 is also expressed on regulatory T cells, which are immunosuppressive, its precise net effect depends on which cell populations are most strongly engaged in a given tissue context.
