This mechanism combines two distinct immune-engaging elements in a single fusion protein: an antibody arm that recognizes specific segments of the T-cell receptor, called Vβ6 and Vβ10, and a linked interleukin-2 (IL-2) domain that signals through the IL-2 receptor. T cells use their receptors to recognize antigens, and each receptor is built from variable gene segments; targeting particular Vβ families allows selective engagement of a defined subset of T cells rather than the entire T-cell population. The attached IL-2 portion then delivers a growth and activation signal to those engaged cells, and to natural killer cells, which also carry IL-2 receptor components. The rationale is to achieve targeted expansion and activation of a T-cell subset already primed to react to relevant antigens, producing more potent and durable killing of abnormal or infected cells, while limiting the broad, unselective immune stimulation associated with administering IL-2 alone. This general strategy is relevant to cancers where reinvigorating cytotoxic T-cell and natural killer cell activity can improve tumor control, and conceptually to other settings where selective expansion of defined lymphocyte populations is desirable.
