Trace amine-associated receptor 1 (TAAR1) is a G-protein-coupled receptor found mainly in brain regions that regulate mood, cognition, and reward, as well as in some peripheral tissues. It is activated not only by classical neurotransmitters but by trace amines, molecules present in the body at very low concentrations that modulate dopamine, serotonin, and glutamate signaling. Because TAAR1 sits upstream of these major neurotransmitter systems, agonists that activate it can dampen dopaminergic overactivity and adjust serotonergic and glutamatergic tone without directly blocking dopamine receptors, which is the mechanism used by most conventional antipsychotic medicines. This distinction matters because dopamine receptor blockade, while effective against psychotic symptoms, is also responsible for movement disorders and metabolic disturbances commonly seen with older therapies. TAAR1 agonism is being explored as a way to influence the same circuits implicated in schizophrenia and related psychotic disorders through a different entry point, with the therapeutic rationale that indirect modulation of dopamine release may achieve symptom control while producing a different tolerability profile. Interest in this target also extends to conditions involving disrupted reward and arousal circuits, such as substance use disorders, given TAAR1's role in regulating dopaminergic neuron firing.
