First-in-class oral STAT6 degrader

STAT6 is a transcription factor that carries signals from two cytokines, interleukin-4 and interleukin-13, from the cell surface into the nucleus, where it switches on genes responsible for allergic-type immune responses. When interleukin-4 or interleukin-13 bind their receptors, an associated kinase enzyme adds a phosphate group to STAT6, causing it to pair up with another STAT6 molecule and move into the nucleus to activate transcription. This pathway drives production of immunoglobulin E antibodies, recruitment of eosinophils, and mucus overproduction, processes central to conditions such as atopic dermatitis, asthma, and other allergic or eosinophilic diseases. Rather than blocking the receptor or the upstream kinase, a STAT6 degrader works by eliminating the STAT6 protein itself, using a small molecule that recruits the cell's own protein-disposal machinery to tag STAT6 for destruction. This approach aims to shut down interleukin-4 and interleukin-13 signaling more completely and durably than agents that only inhibit STAT6's enzymatic activation, while sparing other signaling routes that share the same upstream kinases. Because the molecule is taken orally rather than injected, it also offers a different route of administration compared to biologic therapies that target the same pathway upstream, at the receptor or cytokine level.
