RXR (retinoid X receptor) is a nuclear hormone receptor that regulates gene transcription by pairing with partner receptors as a heterodimer. One of its partners, Nurr1, is a nuclear receptor essential for the development and ongoing survival of dopamine-producing neurons in the midbrain. Because Nurr1 itself lacks a well-defined pocket for small-molecule activation, one strategy is to activate the RXR side of the RXR/Nurr1 pair, which can switch on the same protective gene programs that Nurr1 normally drives. This approach is of particular interest in Parkinson's disease and other conditions marked by progressive loss of dopaminergic neurons, where boosting neuroprotective and pro-survival gene expression in these cells could slow degeneration or support neuronal resilience. More broadly, RXR modulation is a way to influence transcriptional programs governed by several nuclear receptor pairings, since RXR partners not only with Nurr1 but with receptors involved in lipid, retinoid, and thyroid hormone signaling. Selectively engaging the RXR/Nurr1 axis, rather than RXR broadly, is intended to concentrate therapeutic effect on dopaminergic neuron biology while limiting the wider hormonal signaling changes that non-selective RXR activation can produce elsewhere in the body.
