RAC1 is a small GTPase, a molecular switch protein that cycles between an active, GTP-bound state and an inactive, GDP-bound state. In its active form, RAC1 organizes the actin cytoskeleton, the internal scaffolding that gives cells their shape and allows them to move, and it also drives production of reactive oxygen species and cell proliferation signals. Because RAC1 sits downstream of growth factor receptors and adhesion molecules, it is a converging point for signals that push cells to migrate, invade surrounding tissue, and survive stress. In many solid tumors, RAC1 activity is abnormally elevated, contributing to invasive growth and to resistance against other targeted therapies. Inhibiting RAC1 aims to blunt this migratory and pro-survival signaling, potentially slowing tumor spread and restoring sensitivity to other treatments. Beyond oncology, RAC1 has roles in immune cell trafficking and vascular biology, making it a mechanism of broader interest wherever excessive cell movement or oxidative signaling drives disease. Because RAC1 is a GTPase rather than a classical enzyme with a well-defined catalytic pocket, developing selective small-molecule inhibitors against it has historically been technically demanding, which shapes how such compounds are designed and evaluated.
