PTPN2 and PTPN1 (also called TC-PTP and PTP1B) are intracellular enzymes belonging to the protein tyrosine phosphatase family. Their normal role is to remove phosphate groups from signaling proteins, acting as brakes on pathways activated by cytokines, interferons, and insulin-like growth factors. In cancer immunology, tumors and the immune cells trying to attack them are subject to this same braking activity: PTPN2 in particular dampens interferon-gamma signaling in T cells and tumor cells, limiting how strongly the immune system responds. By inhibiting PTPN2 and, given the close structural similarity of their catalytic sites, often PTPN1 as well, a small molecule can sustain and amplify cytokine signaling, making tumors more visible to the immune system and making T cells more responsive to activation. This rationale places such inhibitors within the broader immuno-oncology strategy of removing intrinsic checkpoints on immune signaling rather than only blocking cell-surface checkpoint receptors. Beyond oncology, PTPN1 has separately been studied for its role in dampening insulin receptor signaling, linking this enzyme family to metabolic disease as well. The dual profile of these inhibitors reflects target biology rather than deliberate polypharmacology design, since separating the two enzymes pharmacologically is difficult.
