PRMT1 (protein arginine methyltransferase 1) is an enzyme that adds methyl groups to arginine residues on proteins, a modification called asymmetric dimethylation. It is the most active member of the protein arginine methyltransferase family and modifies histones—the proteins around which DNA is packaged—as well as many non-histone proteins involved in gene transcription, RNA processing, and the cellular response to DNA damage. By methylating histone tails, PRMT1 helps set the chromatin state that determines which genes are turned on or off. In several cancers, PRMT1 activity is elevated and contributes to programs that support tumor cell proliferation and survival, partly through effects on transcription factors and splicing regulators. Small-molecule inhibitors of PRMT1 aim to reduce this excess methylation activity, thereby disrupting the transcriptional and RNA-processing programs that support malignant growth. Beyond oncology, arginine methylation by PRMT1 also influences broader cell signaling networks, so this mechanism has been of interest wherever dysregulated methyltransferase activity is thought to drive disease. Because PRMT1 acts on many different protein substrates rather than a single pathway, inhibiting it is expected to have pleiotropic effects, which is both the rationale for its therapeutic interest and a source of potential on-target consequences elsewhere in normal tissue.
