PPAR delta and PPAR gamma are members of the peroxisome proliferator-activated receptor family, nuclear hormone receptors that act as ligand-activated transcription factors. When bound by a small molecule or natural fatty-acid-derived ligand, they partner with the retinoid X receptor and bind specific DNA sequences to switch on or off genes governing lipid and glucose metabolism. PPAR gamma is enriched in adipose tissue and is the principal driver of adipocyte differentiation and insulin sensitization, while PPAR delta is more broadly expressed and primarily promotes fatty acid oxidation and cellular energy use, particularly in skeletal muscle and liver. A dual agonist engages both receptors simultaneously, aiming to combine gamma-driven improvements in insulin sensitivity with delta-driven increases in fat burning and energy expenditure, an approach intended to produce a broader metabolic benefit than activating either receptor alone. This mechanism is of interest across metabolic diseases characterized by insulin resistance and disordered lipid handling, including type 2 diabetes and fatty liver disease, and has also been explored in neurodegenerative conditions where impaired brain glucose metabolism and insulin signaling are thought to contribute to disease progression.
