Janus kinases (JAK1, JAK2, JAK3, and TYK2) are enzymes that sit just inside the cell membrane, attached to receptors for cytokines—signaling proteins that coordinate immune responses. When a cytokine binds its receptor, associated JAK enzymes activate, adding phosphate groups to intracellular proteins called STATs, which then travel to the nucleus and switch on genes involved in inflammation and immune cell activity. A 'pan-JAK' inhibitor blocks all four family members rather than a single subtype, providing broad suppression of cytokine signaling because many inflammatory skin conditions are driven by multiple, overlapping cytokine pathways rather than just one.
