Peptides and proteins are difficult to deliver orally because they are large, often charged molecules that are degraded by stomach acid and digestive enzymes, and that cannot easily cross the intestinal lining to reach the bloodstream. Oral macromolecule delivery capsules are engineered devices, sometimes combined with chemical formulation strategies, designed to overcome these barriers so a peptide or protein drug that would normally require injection can instead be swallowed. Approaches in this class generally rely on protecting the payload from gastric degradation until it reaches the intestine, then using a mechanical, chemical, or physical mechanism to push the molecule across or through the intestinal wall, bypassing the normal absorption barrier. The rationale for this technology class is broad: any macromolecule therapy that is currently limited to injection, such as insulin, GLP-1 receptor agonists, or various therapeutic proteins, is a potential candidate for an oral route if the delivery barrier can be solved. Successfully doing so could improve convenience, reduce the burden of chronic injections, and expand access to biologic-class medicines. This makes the platform relevant across many disease areas, including diabetes, metabolic disease, and other chronic conditions where injectable biologics are standard therapy but where an oral alternative would be preferred by patients and prescribers.
