NLRP3 (NOD-, LRR- and pyrin domain-containing protein 3) is a cytoplasmic sensor protein that forms part of the innate immune system, the body's first line of defense against infection and tissue damage. Upon detecting danger signals—such as microbial products, crystalline material, or markers of cellular stress—NLRP3 assembles with adaptor and enzyme partners into a multiprotein complex called the inflammasome. This complex activates caspase-1, an enzyme that cleaves precursor forms of the inflammatory messengers interleukin-1beta and interleukin-18 into their active forms and triggers a form of programmed inflammatory cell death called pyroptosis. While this response is protective during acute infection, inappropriate or persistent NLRP3 activation is implicated in a broad range of chronic inflammatory and metabolic conditions, including gout, atherosclerosis, neurodegenerative disease, and inflammatory disorders of the lung, liver, and skin. Small-molecule inhibitors that block NLRP3 assembly or activation aim to dampen this specific inflammatory pathway upstream of interleukin-1beta production, offering a more targeted alternative to broader immunosuppression. Because NLRP3 sits at a convergence point for many types of cellular stress, it is considered a mechanistically attractive node for treating diseases where sterile, non-infectious inflammation drives tissue damage.
