mGlu4 (metabotropic glutamate receptor subtype 4) is a G-protein-coupled receptor belonging to the group III family of metabotropic glutamate receptors. It is located mainly on presynaptic nerve terminals, where its activation by the neurotransmitter glutamate reduces further release of glutamate and other transmitters, acting as a feedback brake on synaptic signaling. Because it is coupled to inhibitory G proteins, engaging mGlu4 generally dampens neuronal excitability at the synapses where it resides. This receptor is enriched in brain circuits that regulate movement, including basal ganglia pathways implicated in Parkinson's disease, and it also has roles in circuits governing anxiety and pain processing. A positive allosteric modulator (PAM) is a molecule that binds to a site distinct from where glutamate itself binds and enhances the receptor's response to that natural ligand, rather than activating the receptor on its own. This approach aims to restore or amplify mGlu4 signaling selectively where and when glutamate is already present, offering a way to correct dysregulated circuit activity in movement and mood disorders while attempting to preserve the receptor's normal, activity-dependent pattern of function.
