LANCL2 activator

LANCL2 (LanC-like protein 2) is an intracellular signaling protein expressed widely in immune and epithelial cells. It functions as a receptor for small endogenous and dietary molecules, most notably abscisic acid, a compound also found in plants that has immunomodulatory effects in mammals. Engaging LANCL2 with an activating ligand triggers signaling that dampens pro-inflammatory immune responses: it favors the development and activity of regulatory T cells, which suppress excessive immune activation, while restraining pathways that drive production of inflammatory cytokines. Because this shifts the local immune balance toward tolerance rather than attack, LANCL2 activation is of particular interest in chronic inflammatory diseases of the gastrointestinal tract, such as ulcerative colitis and Crohn's disease, where the mucosal immune system is persistently overactive against the gut lining and its resident microbes. Unlike therapies that broadly suppress immune function, LANCL2 activators are studied as agents that recalibrate immune signaling locally, with the goal of reducing tissue damage while preserving the capacity to respond to genuine infectious threats. Oral small molecules that activate LANCL2 are designed to act largely within the gut, aligning drug exposure with the site of disease.
