This bispecific antibody is designed to block two receptors that both drive so-called type 2 inflammation, the immune pattern behind allergic diseases such as asthma, atopic dermatitis, and chronic rhinosinusitis. IL-4 receptor alpha (IL-4Rα) is a shared subunit used by two cytokines, interleukin-4 and interleukin-13, which together promote IgE antibody production, mucus secretion, and recruitment of eosinophils (a white blood cell subtype associated with allergic tissue damage). ST2 is the receptor for interleukin-33, an "alarmin" released by damaged or stressed epithelial tissue that alerts the immune system early in the inflammatory cascade, activating innate lymphoid cells and mast cells that reinforce the type 2 response. By neutralizing both receptors with a single molecule, this approach aims to interrupt the pathway at two points: upstream, where tissue damage first triggers alarm signaling, and downstream, where cytokines execute the allergic response. This dual blockade may offer broader control of inflammation than targeting either receptor alone, particularly in patients whose disease is driven by overlapping or redundant type 2 signals. The rationale reflects a broader trend in immunology of combining pathway nodes to address disease heterogeneity, where single-target therapies sometimes leave residual inflammation unaddressed.
