Interleukin-2 receptor alpha (IL2RA, also called CD25) is one of three subunits that assemble into the high-affinity receptor for interleukin-2 (IL-2), a cytokine that drives proliferation and survival of T lymphocytes. CD25 itself does not transmit a signal; its role is to bind IL-2 with high affinity and hand it to the signaling beta and gamma chains. CD25 is expressed constitutively on regulatory T cells (Tregs), the subset that restrains immune responses, and is induced transiently on conventional T cells once they become activated. Because of this expression pattern, agents directed at CD25 can be designed for opposite therapeutic purposes: blocking IL-2 binding dampens immune activation and is useful where the goal is suppressing an unwanted immune response, such as organ transplant rejection or autoimmune disease. Conversely, depleting or otherwise eliminating CD25-high cells can selectively reduce Tregs, releasing suppression on tumor-reactive T cells and enhancing antitumor immunity. Antibody-based agents against CD25 exploit these possibilities, and design choices such as whether the antibody blocks IL-2 binding or instead engages immune effector mechanisms to kill the target cell determine which of these opposing effects predominates. This dual therapeutic logic makes CD25 a target of interest across transplantation, autoimmunity, and oncology.
