Interleukin-17 receptor A (IL-17RA) is a cell-surface receptor expressed on epithelial cells, fibroblasts, and various immune cells. It functions as the shared signaling subunit for a family of related cytokines, including IL-17A, IL-17F, and the IL-17A/F heterodimer, each of which pairs IL-17RA with a second receptor chain to trigger downstream signaling. This pathway drives production of inflammatory mediators that recruit neutrophils and amplify tissue inflammation, and it is markedly overactive in immune-mediated skin and joint diseases such as psoriasis and psoriatic arthritis. Blocking IL-17RA directly, rather than targeting a single cytokine such as IL-17A, interrupts signaling from multiple IL-17 family members at once, since they converge on this common receptor chain. This broader blockade can produce more complete suppression of the downstream inflammatory program than neutralizing one ligand alone. Anti-IL-17RA monoclonal antibodies bind the receptor and prevent it from engaging its cytokine partners, thereby dampening the inflammatory cascade that leads to visible plaques, joint swelling, and other tissue damage. This mechanism is relevant broadly across IL-17-driven autoimmune and inflammatory conditions, where excessive cytokine signaling through this receptor sustains chronic tissue injury.
