Anti-IgE monoclonal antibody

Immunoglobulin E (IgE) is an antibody class produced by plasma cells that plays a central role in allergic disease. Under normal circumstances IgE helps defend against parasites, but in susceptible individuals it is generated against otherwise harmless substances such as pollen, dust mite proteins, or certain foods. Once produced, IgE binds tightly to a high-affinity receptor found on the surface of mast cells and basophils, effectively arming these cells. When the allergen is encountered again, it cross-links the receptor-bound IgE, triggering the release of histamine and other inflammatory mediators that produce the symptoms of allergic reactions: airway constriction, swelling, itching, and mucus production. Anti-IgE monoclonal antibodies are designed to bind circulating free IgE before it attaches to mast cells and basophils, reducing the pool of IgE available to sensitize these cells and lowering receptor density over time. This approach addresses a shared upstream driver of multiple allergic conditions, including asthma, chronic hives, and nasal polyps, rather than blocking a single downstream mediator. By intervening early in the allergic cascade, this mechanism aims to reduce the frequency and severity of allergic flares across a range of IgE-mediated diseases.
